|Test||HEREDITARY NONPOLYPOSIS COLORECTAL CANCER (HNPCC) SCREEN, TISSUE|
|Methodology||Polymerase Chain Reaction (PCR is utilized pursuant to a license agreement with Roche Molecular Systems, Inc.)
Avidin-Biotin Complex Method Immunoperoxidase Stain
|Description||Identification of individuals at high risk for having a diagnosis of HNPCC.
HNPCC screen will always include: microsatellite instability, tumor and slide review for ancillary tests, MLH1, MSH2, MSH6 PMS2 stains.
|Specimen||"Molecular Genetics - Inherited Cancer Syndromes Patient Information Sheet" is required for all orders.
Send paraffin-embedded tissue block or 1 slide stained with
hematoxylin-and-eosin and 10 unstained, nonbaked slides (5 slides with 5-Ám thick sections and 5 slides with 10-Ám thick sections) of the tumor tissue.
Note: Sections should contain both tumor and normal tissue.
|Reference Values||An interpretive report will be provided.
The report will include specimen information, assay information, and interpretation of test results.
Microsatellite stable (MSS) is reported when none of the markers
demonstrate MSI, MSI-L when MSI occurs in <30% of microsatellites examined, and MSI-H when MSI occurs in >30% of microsatellites examined. MLH1, MSH2, MSH6, and PMS2 protein expression, as assessed by IHC, is normally expressed in nonmalignant tissue.
